• This contract is part of the VERTICAL research project (Mechanistic characterization of novel host and viral factors for therapeutic interventions targeting transplacental transmission of Flaviviruses), funded by PEPR Maladies Emerging Infectious Diseases (EIM), and in particular its Work-Package 4 (WP4). The VERTICAL project aims to elucidate the molecular and cellular mechanisms (host and viral factors) which govern the transmission of Flaviviruses (such as viruses Zika, Usutu, Dengue or West Nile) from mother to fetus during pregnancy. The major challenge is to identify new therapeutic targets (in particular to evaluate the impact of placental extracellular vesicles) to prevent infections. congenital and protect fetal development. This purchase is specifically dedicated to the operational implementation of WP4. This component of the project aims to: - - - Develop and operate a high-throughput cellular imaging (HCS) technological platform to model viral infection at the mother-child interface. Characterize the biological impact of placental extracellular vesicles (EVs) on viral transmission. Identify, through pharmacological screening, compounds capable of inhibiting viral replication in complex human models. The success of this project relies on the service provider's ability to combine cutting-edge expertise in automated imaging with the handling of highly sensitive human biological samples. (including fresh placental explants). This service requires strong technical integration with the teams of the Inserm Infinity institute, project leader, in order to transform ex vivo biological models into quantitative screening data. and reproducible.
